MPGŪ Guanidine Direct mRNA Purification Kit - Phenol Free
For isolation of mRNA from cells or animal tissue containing high levels of RNase.

The MPGŪ Direct mRNA Purification Kits are designed specifically for the isolation of pure, intact mRNA directly from cells and animal or plant tissue. The isolation takes 15 minutes or less and eliminates the need for total RNA isolations, oligo(dT) columns, phenol-chloroform extractions and long overnight centrifugations through CsCl gradients. The kits contain all of the reagents necessary to isolate and purify high quality mRNA up to 1.0 gram of tissue or 1 x 108 cells. A typical range of 30-65 µg of mRNA can be purified per kit. Increased yields are obtained from pooling multiple rounds of mRNA isolated from the same tissue, using the same particles.

The key component of the kits, MPGŪ Streptavidin Complex, provides greater surface area than other magnetic particles, resulting in greater oligo(dT) binding and, therefore, generating greater mRNA capture. Typical yields of 3-7 µg/mg of solid phase are achieved from as little as 0.05 grams of tissue (mouse liver). The messenger RNA is captured directly from tissue or cell homogenates on oligo(dT)25 - bound MPGŪ Streptavidin and released in water with purities >1.8 as measured by A260/A280.

The mRNA purified by these kits is ready for direct use in: cDNA library construction; Northern blotting; in vitro translation; S1 nuclease analysis; sequencing; dot blot hybridization; RNA binding protein isolation; subtractive hybridization; cloning; and RT-PCR.

mRNA yield is dependent on the origin of the tissue.

mRNA was isolated from fresh mouse tissues homogenized in Tissue Extraction/Hybridization Buffer (0.05 g/ml). 1 ml homogenates were incubated with 1 mg MPGŪ Streptavidin-oligo(dT)25 Complex. (following protocol 5.2)
