The Solid Phase Oligo COUPLE-IT Kit is the first kit available for convenient coupling of primary amine-labeled oligonucleotides and other primary amine-labeled nucleic acids to a magnetic solid support (Magnetic Porous Glass - MPG™). The kit contains all of the reagents and magnetic solid support needed to easily carry out 2 optimal coupling reactions (10 mg of solid support per reaction). The resulting product is a reusable oligo-coupled magnetic solid support which can be stored for future use.
The kit protocol is simple to follow and optimized to remove the guess work from an otherwise complex procedure. The amine-modified oligo or nucleic acid to be coupled to the solid support can be: obtained from any oligo synthesis facility; generated using standardized chemical labeling procedures; or generated by incorporation of aminoallyl-dUTP or -UTP using DNA or RNA polymerase respectively.
The coupled product can be used in solid phase nucleic acid-based applications including: in vitro translation; subtractive hybridization; purification of specific messages or RNA species; isolation of specific DNA molecules; hybridization-based assays; and purification and/or identification of nucleic acid binding proteins. Coupling with the magnetic solid support further enhances the efficiency and convenience of further applications by enabling rapid, single tube assays and in most cases eliminating the need for centrifugation and column chromatography.
The kit also works with proteins and peptides by simply substituting them in molar equivalence for the nucleic acid in the supplied protocol.
