Researcher: I have been using PureBiotech LLC's Solidscript™ Solid Phase cDNA Synthesis Kit to study gene expression. I now want to isolate specific rare messenger RNAs using complementary probes. Can I still use this kit?

TechTALKer:Yes. The first part of the SOLIDscript™ protocol is mRNA isolation. The biotinylated oligo (dT)25 probe contained in the kit is first bound to the MPG® Streptavidin particles before hybridization to the poly-A RNA. By simply replacing this biotinylated oligo (dT)25 probe with any 5'-biotinylated gene specific oligo you can capture a complementary sequence. In fact, transcriptional promoters such as the E. coli T7 promoter have been successfully integrated into the probe sequence resulting in reusable, immobilized in vitro transcription systems. In addition, restriction sites can also be designed to enable subsequent cloning of cDNAs.
