Researcher: There are many different ratios and pH of phenol:chloroform some with isoamyl alcohol. How do I choose the correct formulations?

TechTALKer:The ratios and pH of phenol solutions is application dependent. Removal of proteins from preparations of DNA is accomplished by using phenol:chloroform:isoamyl alcohol (25:24:1), pH 7.8-8.0. The chloroform denatures proteins and assists the partitioning of the organic and aqueous phases. The isoamyl alcohol reduces foaming. RNA isolations require an acidic pH to partition the genomic DNA into the organic phase leaving only the RNA in the top aqueous phase. The recommended formulation for RNA isolation is phenol:chloroform:isoamyl alcohol (125:24:1), pH < 5.0.
